Execution of Visium Spatial Gene Expression (Visium), RNAscope, and related software analysis software - Visium enables RNA sequencing to be performed on tissue sections, thereby maintaining anatomical orientation. This technology combines throughput sequencing with high-resolution microscope to capture the expression pattern in tissue sections, down to a resolution of 1-10 cells. RNAscope is a novel in situ hybridization technique with a special probe designed to amplify target specific signals without background noise from non-specific signals. This technology is sensitive enough to detect a single transcript in a cell.

Image and statistical analysis, using a variety of programs and software that include Keyence microscope and imaging programs; Prism and/or Sigma Plot; R-Swirl; ImageJ; Photoshop adobe and illustrator; Excel and Powerpoint

Preparation and isolation of protein, RNA, and DNA from cell cultures, fresh tissue, frozen tissue, and formalin fixed paraffin embedded tissue - these specimens will undergo morphologic, proteomic, and genomic analysis.

Initiation and maintenance of human and mammalian corneal epithelial cells, which includes the construction of organotypic corneal epithelial models - these cells and models will be used to replicate epithelial wounds for subsequent protein and genomic analysis.

Execute western blots and subsequent analysis with either SDS-PAGE gels or Protein Simple Wes Western Blot technology.

Anesthetization, handling, wounding, imaging, and analysis of genetic mouse models - mice will be used for in vivo corneal wounding experiments.
